免疫逃避是T细胞肿瘤免疫治疗所面临的主要障碍之一。肿瘤排斥抗原表达不足会导致固有免疫抵抗和干扰素γ (IFN-γ)诱导的程序性死亡配体1(PD-L1)高表达以进一步诱导免疫抵抗。鉴于此,华东师范大学徐志爱教授和中科院上海药物研究所于海军研究员开发了一种多功能前药纳米囊泡用于Doxy的肿瘤靶向递送。该前药纳米囊泡具有可脱落的聚乙二醇壳层和CRGDK配体,在血液循环中具有较高的稳定性;该纳米药物能够募集T淋巴细胞浸润肿瘤,并显著抑制FN-γ诱导的瘤内PD-L1表达。这一研究可为降低肿瘤免疫抵抗和增强肿瘤免疫治疗提供一种新的靶向策略。
本研究中使用欣博盛小鼠IFN-γ ELISA试剂盒检测了小鼠肿瘤组织中IFN-γ的分泌情况。
