近期,武汉大学黄卫华团队、厦门大学Christian Amatore团队合作在《Nature Nanotechnology》(IF=38.8)上发表题为“Nanosensor detection of reactive oxygen and nitrogen species leakage in frustrated phagocytosis of nanofibres”(纳米电极监测巨噬细胞受挫性吞噬纳米纤维过程中ROS/RNS泄漏)的研究,报告了一种电化学纳米传感器,用于监测和定量表征单个巨噬细胞对玻璃纳米纤维的吞噬作用受阻期间活性物质释放的通量和动态。
机体吸入环境暴露的高长径比惰性纳米纤维(石棉、玻璃纤维、碳纳米管等)后,肺泡巨噬细胞会迅速识别这些异物,并试图将其吞噬。由于纤维状材料的高长径比和难于分解的特性,巨噬细胞难以将其快速清除,从而形成受挫性吞噬作用(frustrated phagocytosis)。在此过程中,巨噬细胞内的NADPH氧化酶(NOX)和诱导型一氧化氮合酶(iNOS)会持续产生活性氧和活性氮(ROS/RNS)消化纤维。由于吞噬腔未封闭,大量产生的ROS/RNS可能会从吞噬口泄漏到胞外,对周围肺组织细胞造成损伤并引发疾病。在过去几十年里,研究人员采用ROS/RNS染色以及活体实验对碳纳米管和石棉等纳米纤维材料的研究均支持这一假设。然而,因缺乏合适的传感器,该过程中各类活性物质的化学组成及其时空动态变化特征仍然未知。
针对受挫性吞噬作用假说,研究团队将铂黑纳米线电化学传感器(SiC@PtNWS)定位在巨噬细胞吞噬口处,从而实现对受挫性吞玻璃纳米纤维的过程中4种主要ROS/RNS(ONOO-,H2O2,NO和NO2-)泄漏通量的实时定量监测。结果表明,在吞噬各阶段,ROS/RNS释放存在明显时间差异。相较于细胞顶部,吞噬口处存在大量ROS/RNS泄漏,且随吞噬进行持续增加直到吞噬口封闭泄漏。NOX和iNOS参与动态调控ROS/RNS的物质组成。相关数据定量展示了单个巨噬细胞受挫性吞噬作用玻璃纳米纤维时ROS/RNS产生的时空特征,从而描绘出巨噬细胞应对惰性高长径纳米材料的动态免疫调节过程。体外共培养和活体实验进一步表明,持续且大量的ROS/RNS泄漏会导致外周细胞严重受损,最终转化为慢性炎症和肺损伤。
