研发背景:
8D3单克隆抗体可识别小鼠CD71蛋白,也被称为转铁蛋白受体抗体1(TfR1)。CD71是分子量大小为170-180kDa的II型同型二聚跨膜糖蛋白,该蛋白表达于增殖细胞、网状细胞和类红细胞前体等细胞的表面。
有研究表明,CD71在调节细胞增殖的过程中发挥作用。另外,CD71还通过内吞作用协助运铁蛋白将铁转运到胞内。 CD71在恶性肿瘤细胞上大量表达,其表达量与癌症的进展、进程相关。由于CD71在恶性肿瘤细胞上的高表达产生的细胞内化能力以及铁对癌细胞增殖的必要性使转铁蛋白受体(TfR1)成为将药物输送到恶性细胞中的关键靶标以及研究热点。相关研究结果显示8D3抗体可在体内消耗CD71 +细胞。
